چکیده مقاله:

The objective of this research is to identify Arabidopsis thaliana genes encoding acid phosphatases induced by phosphate starvation. Multiple alignments of eukaryotic acid phosphatase amino acid sequences led to the classification of these proteins into four groups including purple acid phosphatases (PAPs). Specific primers were degenerated and designed based on conserved sequences of PAPs isolated from plants, fungi and animals. RNA profiles of Pi-fed and Pi-starved A. thaliana roots were compared with two methods established for gene-family-directed differential display of pap genes. Having analyzed the differentially displayed fragments, seven pap encoding cDNA clones were isolated. One of the clones was a trans-splicing product of two genes that encodes an acid phosphatase carrying a zinc-finger domain. Six other clones were predicted to encode secretory phosphatases. Reverse-northern blotting and semi-quantitative RT-PCR revealed distinct expression patterns for each gene under diverse environmental conditions such as Pi starvation, high-salt concentration, cold shock, nitrogen and sulfur deprivation. The presented data can provide some clues for dissecting the possible roles of PAPs in Pi acquisition.

Identification and Expression Analysis of Two Arabidopsis LRR-Protein Encoding Genes Responsive to Some Abiotic Stresses

AbstractTwo Arabidopsis thaliana genes, psr9.2 and psr9.4 appearedto be highly similar to a phosphate-starved induced gene,psr9, isolated from Brassica nigra suspension cells.Sequence analysis classified the encoded polypeptides asmembers of leucine-rich repeat (LRR) proteins superfamily. The sequence of psr9 proteins comprise a unique N-terminalregion e...

Update on Abiotic Stresses in Arabidopsis and Grasses Transcriptional Regulatory Networks in Response to Abiotic Stresses in Arabidopsis and Grasses

Various abiotic stresses such as drought, high salinity, high temperature, and low temperature negatively impact plant growth and productivity of crops. Plants have adapted to respond to these stresses at the molecular, cellular, physiological, and biochemical level, enabling them to survive. Various adverse environmental stresses induce the expression of a variety of genes in many plant specie...

MhYTP1 and MhYTP2 from Apple Confer Tolerance to Multiple Abiotic Stresses in Arabidopsis thaliana

The first YTH domain-containing RNA binding protein (YTP) was found in rat, where it was related to oxidative stress. Unlike characterizations in yeast and animals, functions of plant YTPs are less clear. Malus hupehensis (Pamp.) Rehd. YTP1 and YTP2 (MhYTP1 and MhYTP2) are known to be active in leaf senescence and fruit ripening. However, no research has been published about their roles in stre...

AtHD2D Gene Plays a Role in Plant Growth, Development, and Response to Abiotic Stresses in Arabidopsis thaliana

The histone deacetylases play important roles in the regulation of gene expression and the subsequent control of a number of important biological processes, including those involved in the response to environmental stress. A specific group of histone deacetylase genes, HD2, is present in plants. In Arabidopsis, HD2s include HD2A, HD2B, HD2C, and HD2D. Previous research showed that HD2A, HD2B, a...

Differential expression of genes for response regulators in response to cytokinins and nitrate in Arabidopsis thaliana.

In Arabidopsis thaliana, a number of response regulators are presumably involved in His-Asp phosphorelay signal transduction in response to environmental stimuli, such as phytohormones. Previously, it was shown that expression of a certain set of genes for response regulators are cytokinin- and nitrate-responsive in their mRNA accumulation, under certain growth conditions [Taniguchi et al. (199...
